چکیده

Background: Infections caused by carbapenem-resistant gram-negative bacteria are treated with colonistin as a last resort. However, the increased resistance in recent years is significant for treating multiple drug-resistant infections. Objectives: This study aimed to determine genes mobile colistin (mcr)-1 and mcr-2 colistin-resistant isolates, understand mechanism, help treatment. Methods: Isolates were identified Vitek MS automated system. The antibiotic susceptibility was tested 2 Compact Enteric bacilli isolates stored at -20°C until molecular study. DNA extraction of performed boiling method. Then, polymerase chain reaction (PCR) optimization using specific primers, mcr-1 investigated multiplex PCR Results: About 170 enteric isolate mainly sent from internal medicine (44.7%) neurology (13.5%) services. According species identification, 37.6% Klebsiella pneumonia, 31.7% Serratia marcescens. Based on results, not detected isolates. Conclusions: Increased worrying discovery mcr require urgent precautions, even though did detect mcr-2. New studies investigating new needed mechanism identify resistant

Colistin MIC variability by method for contemporary clinical isolates of multidrug-resistant Gram-negative bacilli.

In vitro evaluation of colistin susceptibility is fraught with complications, due in part to the inherent cationic properties of colistin. In addition, no reference method has been defined against which to compare the results of colistin susceptibility testing. This study systematically evaluated the available methods for colistin MIC testing in two phases. In phase I, colistin MICs were determ...

Preservation of Acquired Colistin Resistance in Gram-Negative Bacteria.

Colistin-resistant mutants were obtained from 17 colistin-susceptible strains of Acinetobacter baumannii, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Escherichia coli. The stability of colistin resistance in these mutants was investigated. Three of four colistin-resistant P. aeruginosa mutants recovered colistin susceptibility in colistin-free medium; however, colistin-susceptible revert...

Effects of Efflux Pump Inhibitors on Colistin Resistance in Multidrug-Resistant Gram-Negative Bacteria.

We tested the effects of various putative efflux pump inhibitors on colistin resistance in multidrug-resistant Gram-negative bacteria. Addition of 10 mg/liter cyanide 3-chlorophenylhydrazone (CCCP) to the test medium could significantly decrease the MICs of colistin-resistant strains. Time-kill assays showed CCCP could reverse colistin resistance and inhibit the regrowth of the resistant subpop...

Intravenous colistin in the treatment of sepsis from multiresistant Gram-negative bacilli in critically ill patients

INTRODUCTION The increasing prevalence of multiresistant Gram-negative strains in intensive care units (ICUs) has recently rekindled interest in colistin, a bactericidal antibiotic that was used in the 1960s for treatment of infections caused by Gram-negative bacilli. We conducted the present observational study to evaluate the efficacy of intravenous colistin in the treatment of critically ill...

Prevalence of Drug Resistance in Nonfermenter Gram-Negative Bacilli

Background and Objectives: Non-fementer gram-negative bacilli (NFGB) are ubiquitous pathogen that has emerged as a major cause of health care associated infections. The aim of this study was to determine the prevalence and antimicrobial susceptibility of   NFGB in an Iranian hospital. Materials and Methods: FromJuly 2005 to November 2006 a total of 257 strains of NFGB including 109 (42.41%)...
